This is a question that has been discussed before I'm sure - I'm using a KW transformer strictly for lighting - what 2 posts are best for wiring my ground and hot wire going to a terminal block??

Use "U" for base/ground/common. Use "A" and/or "B" for variable voltage 6-20 volts lighting circuits.
@Harleylito, what @ADCX Rob stated will work for variable voltage. Since you don't indicate the need for a layout common ground, then you can also make use of the fixed voltages available as marked on the breakout, most typical being 14vac with one wire from C and the other from D:

Thanx so much Rob! Can I use A & U for one set of wires going to lights AND B & U for another set of wires to lights in another area of my layout?

You can, but you don't need to split it up if your are running all lights at the same voltage. "A" & "B" are just two taps off of the exact same source.
If your running two terminal blocks as it sounds, I believe you could line them in in a row and run a jumper wire from the last red to the red contact lug on the second one and do the same with black.
