Is there any reason I can't use the PS2 board from a Railking 2-8-0 to upgrade a dual-motor diesel? I have the capability to change the sound file with the loader, but is there any component difference that would make driving two motors inadvisable?

None at all, the boards are identical.
If you are taking that board from a RailKing Pennsy 2-8-0 and using it in a Reading diesel, that is a conversion in more ways than one.

The board is the same, however there's a bit of wiring to do.
It's not that difficult to do a diesel to diesel transplant, but if the board is in the tender steamer, you just have a bunch of wiring. If it's in the locomotive, it's closer, but still stuff to wire.
I've done the reverse, using a diesel kit to upgrade a steam switcher, to get the 2 couplers.
Wowak, I upgraded a Weaver C630 to PS2, right after the first upgrade kits were released. I don't remember how I tackled the couplers, which may mean that they weren't a problem. To this date, that loco is one of the best running locos I own. I also did a Weaver SD40-2 at that time, and also don't remember any problems with it, other than the upgrade was a bear because of the narrow body.

I think I'd rather have had the tether, adding a coupler connector is a lot easier than wiring up the whole tether. The couplers are easy to get, the tether is more expensive than a coupler.

That sounds like a pretty unique case. In general, if I'm doing a steamer, I'd rather start with the steamer wiring harness, less work. Same if I was starting with a diesel...
