I just got a used PRR Railking E-33 from a hobby shop in Hanover PA today, and upon getting home and firing it up, I found it would not work through DCS. In conventional, it works well and has sounds and all that. When I fire it up through DCS, i can hear the "click" sound and a faint buzz, but nothing happens.
Also, when I turn off the engine, the shutdown sounds only last for a second or two.
What is going on and what do i need to fix?
(It does have the original 9-Volt battery from MTH in it)

Replace the battery to start, if it has the original, it's a 20 year old NiCad!

I tried "charging" the loco for 15 minutes or so, and now it just honks randomly and constantly!
What do I replace the battery with?
A 9V NiMh rechargeable battery, these are the ones I use.

As John mentioned. I use these. Easily obtained from local hardware store.
As well........do not attempt to run engine any more until you install a new battery.
Got a new 9V battery today and popped it in the engine.
DCS still didn't pick it up.
What's the next step? Could this be a blown board?
Quite likely actually. 5V PS/2 boards tend to go suddenly and often. 
Some will last forever, others will crap out just sitting on a shelf.
Try to run it conventionally first no DCS signal present. If it runs with new battery, then go back to DCS. Delete the engine if it is in the remote, then try to readd it. If it does not add, try a Recover engine from the engine menu. Must use TIU ID 1 and I recommend having it on Fixed 1. Only have that engine on a test track. G

I don't use the whole TIU set up-i only use the DCS explorer and the iPhone app.
I will try it in conventional with the new battery though.
@GGG and @gunrunnerjohn , it didn't work with the new battery. It ran awkwardly in conventional, but the horn is constantly on. Should i just get a new board for it?
Well, a new board would probably fix it, but maybe it's the environment. Have you tried it on a short test track with nothing else connected? Have you tried a pure sine-wave transformer?
Sounds like a short somewhere (buzz), constant horn. Could be wiring, could be a failing 5V (likely). Turn over to a tech. Ultimately it can be repaired with a PS-32 board if it is not something else. G
