what is the best way to hold down styrofoam on your layout glue or screws and what do you use to cover the seams

Are you talking rigid foam (pink or blue) or styrofoam (white). If its white, don't use it. If it's rigid foam, use a foam safe adhesive like Loctite PL300 and/or hot melt glue to hold it in place and glue pieces together. To hide the seams use WS foam putty.
also, remember to peal the thin plastic covering from top and bottom.

the pink and blue rigid styrofoam insulation I have purchased at lumber yards always had a very thin film covering the top and bottom surfaces which had to be pealed off to be glued and painted

I used blue and Pin 4x8 sheets. They had Tongue and grove and slid together. Also used some 2x8, same thing. I glued some of mine and some I DID NOT!. In hindsight I would have NOT glued any! It all says put. NO SCREWS OR GLUE! As for seams any I had were either not visible or if they were, due to my cutting, I left as the scenery material on top covers it.
I used 2" 4 x 8 OC Pink rigid insulation board for the mountain area bases and then added rolled up paper for shaping the mountain...all covered in plaster cloth. The adhesive used was the heavy duty liquid nails...no issues and solid hold.
