More information would be helpful including Manufacturer, product number, and power/control setup (conventional, TMCC, or Legacy).  

General troubleshooting:

1.  Make sure smoke switch on engine is on
2.  Add 10-15 drops of smoke fluid to start
3.  18v power to track, start engine
4.  Turn on smoke in Cab1 (press 9) or Legacy controller (press smoking stack icon to cycle low, med, high smoke)
5.  Wait 30 seconds
6.  If the above fails, reprogram the engine using procedure below and repeat #3,4,5.  

(Note that there are additional programming codes for other manufacturer's engines.  Check your manual)
