The non-rush hour drive time is about an hour between the Fairgrounds and the Museum. This year I'd like to attend the museum activities on Wednesday but want to get to the DCS User's Group at the Fairgrounds. Considering the afternoon traffic on Route 30, how much time should I allow for the trip?

Not much more. Give yourself an extra 15 or 20 minutes but try to avoid the rush hour as much as possible.
Check your local maps. If the traffic on 30 gets really bad you may want to head south
on any of the major streets (e.g Route 24 or Sherman) then move west along Route 462 through York center. At Carlisle St. swing left then take a right on Market and you'll be at the fairgrounds.

The last museum presentation (Lionel) is scheduled for 4-5 p.m.
The DCS Group Meeting is scheduled to begin at 6 p.m. (actually 6:15 for meals, followed by the other stuff).
If you leave the museum promptly at 5, you shouldn't miss anything at the DCS meeting unless you needed to have a TIU upgraded (6 p.m.)
More traffic will likely be heading east on 30 at that time of day than heading west.
The Wright's Ferry Bridge, on Rt 30 between Columbia and Wrightsville, is still under contruction through November. Traffic trying to by-pass that bridge ties up the PA 462 bridge just down river. Trying to go from Straburg to the fairgrounds around 5 p.m. on a weekday afternoon without any accidents will probably take longer than one hour.
I recall doing the 462 thing once when there was 30 construction years ago (maybe 5-7 years back?). My impression of the trip was that there were lots and lots of stop signs, turns to continue along the route, etc. A fair amount was through what I consider somewhat densely populated neighborhoods.
I'm sure if there is a road closing accident on 30 it's a better choice, but I wouldn't think it a viable alternative to quickly get from Strasburg to York.
I was considering doing the same thing Bob suggests this time (Museum --> DCS meeting), but I'm starting to second guess the practicality of it.

Thanks all. Great info and good suggestions. Since I'm one of the guys who does the DCS upgrades, I want to be at the White Rose NLT 5:30 so to play it safe, I think I'll leave the Museum after the Weaver presentation.
