I got a Northern Pacific 2 Bay Hopper in the mail yesterday and one truck was off of the car.  The silver truck screw was sheared off on one of the trucks.  It is a Kline 6-21344 NP hopper.  Any one known of a source for these screws?

Start with Brasseur Electric Trains, 989-793-2509. I think they ended up with most of the K-line parts.  The other thing to do is provide a technical description of the screw, size, pitch, overall length, shoulder diameter and length, and someone might be able to match it to a Lionel screw.  Lionel used a lot of special screws, some look just like the one you are looking for.
