Can someone explain in simple terms what the difference is in these two controllers? Are all of the building blocks in the Lionel system interchangeable?

One is the remote only, the other is the remote and command base. You need both the remote and command base for the system to work.
I am pretty sure the ones you referenced are obsolete and have been replaced by the Cab-1L/Base-1L #6-37147 for the complete setup, #6-37155 remote only and #6-37156 base only.
The older ones may be available used, but I would recommend the new Cab/Base-1L system, new with full warranty. There may be a time when the older ones will not be able to be repaired anymore, if we are not there already?
