I had a similar issue with a PTC car. After replacing the board set and loading the correct sound file, I got no passenger station announcements. I did factory and feature resets and no change.
After several calls to MTH, someone suggested doing a conventional reset with a transformer, one whistle and five bells. Darned if that didn't get it's mind right, and everything started working.

I do not understand what you mean.
I believe Ben is referring to what is also known as "Auto Mode" where the station stops (actual locations on your track) can be programmed into the engine and it will then stop where you program it to.
(technically, you put it in "learn mode" while programming and then set it for "auto mode" once programmed)

Try the conventional reset and see if it changes anything. You have to get the timing right and get the two toots of the horn indicating the reset happened. If you don't get the horn, try it again. Here's the text from the manual on the reset procedure.
To override the settings you currently have assigned to the engine and reset it to it's factory defaults, while in Neutral tap the Horn button quickly once, followed by five quick taps of the Bell button, allowing approximately ½ second to lapse between each quick button press. Two horn blasts will indicate that the engine has made the change.
