I have a RailKing B6 0-6-0 and my friend has a Lionel Corporation Tinplate 255E 2-4-2 which have been run on the store's display layout to a point where the flanges on the drivers are worn to a knife edge shape, and so now keep derailing on the layout's O-36 FastTrack curves.  Can we replace these wheels ourselves, or do these locomotives have to go to MTH for repair (if possible)?

Try an MTH authorized repair center first and see what they say about replacing the wheels. The wheels are not easy to get off of the original Lionel 225E steam engine and would think that that the reproduction series engine would be similar. You need a wheel puller or miniture gear puller to reduce damage to the wheel shafts on the original 225E.
