I see these designations from time to time on rolling stock boxes. I'm building a 3rail layout with DCS control. Is there a difference? Can I run either ? Confussed ? Also, what is different about DCS trucks if they are just freight cars ?

The 2-rai cars are made for 2-rail track and are insulated from each other as well as having smaller flanges (as 2-rail track has much smaller rails) and likely smaller, scale-sized couplers. They won't run well on your 3-rail track, or couple easily (if at all) to your existing 3-rail rolling stock.
I suspect the 'DCS' refers to "diecast" metal trucks.

DCS in this case stands for Die-Cast Sprung trucks.
Yardtrain: I understand your confusion. That is the first time ever I saw a box marked liked that. Someone at that company should have thought about it as to not cause confusion to the less experienced.
Thank you for the detailed explanation! I always wondered what the difference was between 2 rail and 3 rail trucks if they were only on freight cars. You saved me money from just taking a chance and finding out the hard way.
Amazed me. Our layout has Gargraves track and Ross switches. All 072 (or a bit more). I have a couple of cars with 2 rail trucks and wheels and Kadee couplers, and they seem to run okay. My original plan was to change the wheels and/or the trucks, but… so far so good. It surprised me. Of course, this may not be the case for everyone.
