I have a MTH 20-3364-1 J 611. I have downloaded the sound file and when I attempt to download the file to the engine the TIU will not recognize the engine. I have reset the engine and with the remote the TIU sees the engine. Any suggestions?

You need to state exactly what you're attempting to do, exactly what steps you're taking to do it and exactly what happens at each step.
The sound file is the correct one for the engine, I have the DCS downloaded and I have done this in the past with no problems though it has been a while. I connect thing up as the list indicates, I am using the serial port. The program picks up com3 on the TIU then it comes up that the engine cannot be found. It tries 3 time and the window pops up.
You are using a programming or track with no other engines on powered tracks.
Can you still add the engine to the dcs remote?
And I guess we need to know what version of the loader are you using.

Yes I reset the engine and added it back into the remote and it operated correctly

I can understand now why to want to try a reload of the sound file. A poor track signal or range problem usually has the whistle constantly blowing until it picks up a good signal.
I don't know why the loader is not finding the engine . I don't think it's a track signal problem but perhaps a cable problem? (I'm just guessing) Don't rush out and buy a new cable. Maybe pull and reseat it.
I will try that tomorrow and I will try a TIU with a USB plug also. I will letyou know the results
I used a newer TIU and USB TO USB cord and everything loaded. The original TIU was one of the first ones.
