None that I know of (and I spent a good amount of time in and around Charlotte over the Christmas holiday). There used to be a couple of decent train stores there--Charlotte Electric Trains, for example--but there's little or nothing there now. My brother-in-law, a fellow train enthusiast (Large Scale) lives in the Charlotte area, so if there was a good train store around he likely would know about it.
Isn't the new Lionel headquarters and store near Charlotte?

NONE in Charlotte. The many that were there are all long gone.
The Little Choo Choo Shop in Spencer is the closest.
The next closest is the Train Loft in Winston-Salem which has a good stock but high prices.

The Lionel store is cool to go see but the stock of scale cars is not good. They have O-27 sized cars and ready to run sets in stock all over the place. The prices are full retail.
The nice thing about the choo choo barn is it is across the street from Spencer Railroad Museum Grounds. A GREAT place to check out the old Southern Shops and restored roundhouse.
BTW - Train Loft is also worth the stop,

I plan to visit the Spencer area and Little Choo-Choo Shop (Choo-Choo Barn is in Strasburg, PA) the next time I'm in NC. We planned to visit there the Saturday after Christmas, but it was closed.
Inquiries about Charlotte train shops come up often on this forum. That area really needs a good train store. Definitely a pity that a city that size and with its railroading history doesn't have a decent train shop.

If you head west of Charlotte, Chuga Chuga Choo Choo has reopened in downtown Kings Mountain. You never know what you'll find in there. I got a great deal on some hardly used FasTrack a few weeks ago. Tons of trains and stuff, mostly O gauge.
