There's a store selling the MTH re482 "SBB Cargo" locomotives with the description "first series, with round front". So that raises the question if there was a second production run done (third?) and if there are other differences between the runs? Does anyone have more information on this?

These locomotives (well, pretty much all the European locos) are re-cataloged each year so there is a very good chance that there have second or even third runs. The "round front" description seems to just be a flub on the part of the description.
There are detail differences between roadnames, but the main body shell stays the same.

Actually I did find there was a difference in bodyshell between the series, there was a thread with pictures: https://ogrforum.com/topic/mth-special-run
Oh well, I was having doubts about the repainting thing anyway
MTH produced both the Traxx AC1 and AC2 body styles; unfortunately early on their catalogue descriptions were backwards. I haven't checked to see if MTH has corrected that -- my observation has been that they typically don't. You can do a search here on the forum for "Traxx AC1 AC2" and find the previous post on this topic.
The AC2 has the downward curve at the ends of the sideframes.
